The Triggering Receptor Expressed on Myeloid Cells 2 (TREM-2), is a protein encoded by the TREM2 gene in the human body. TREM-2 protein is mainly expressed in immune cells in various tissues. It is increasingly recognized as an important receptor in different inflammatory diseases and may have the potential as a therapeutic target in the future.TREM-2 binds to the homo-dimer DAP12, leading to the phosphorylation of the Immunoreceptor Tyrosine-based Activation Motif (ITAM) on it by Tyrosine kinases. This then recruits Syk tyrosine kinase, which in turn, along with GRB2/SOS1 and DOK3, activates PLCγ, further inducing anti-inflammatory cytokine responses by elevating intracellular Ca2+ through IP3 activation and inducing cellular cytokine responses through cell rearrangement and phagocytosis.TREM2 Reporter Jurkat Cell Line is a luciferase reporter cell line constructed based on the PLC signaling pathway. Through pathway activation, it activates the expression of Firefly luciferase. The Luciferase readout represents the activation effect of the pathway, therefore, it can be used for the in vitro evaluation of TREM2-related drugs.
