TREM-1 (Triggering Receptor Expressed on Myeloid cells 1) is a protein encoded by the TREM1 gene, expressed on bone marrow cells. Inflammatory responses mediated by monocytes/macrophages and neutrophils can be triggered by various receptors including G protein-coupled receptors (e.g. FPR1), Fc receptors, CD14, Toll-like receptors (TLR; e.g. TLR4), and cytokine receptors. TREM-1 amplifies toll-like receptor-induced inflammation by further increasing the production of pro-inflammatory cytokines.TREM-1 binds to homodimer DAP12, leading to phosphorylation of its immunoreceptor tyrosine-based activation motifs (ITAM) by tyrosine kinases. This recruits spleen tyrosine kinase SYK and ZAP70, activating the PLC pathway, increasing intracellular Ca2+ levels, inducing anti-inflammatory cytokine responses, and activating transcription factors.The TREM1 Reporter Jurkat Cell Line is a luciferase reporter cell line constructed based on the PLC signaling pathway. Activation of transcription factors leads to the expression of luciferase. Luciferase readings represent the activation effect of the signaling pathway, making it suitable for evaluating the in vitro effects of TREM1-related drugs.
