THLE-2 cells are immortalized human adult liver epithelial cells modified with recombinant simian virus 40 large T antigen virus. These cells have undergone more than 100 population doublings and exhibit a near-diploid karyotype. They have been found to be non-tumorigenic when injected into athymic nude mice. Notably, THLE-2 cells do not express alpha-fetoprotein.
In the early passage, THLE-2 expresses cytokeratin 18 and albumin. However, higher-passage cells in logarithmic-phase growth also express cytokeratin 19. THLE-2 cells possibly possess functional cytochrome P450 pathways, as they are able to metabolize benzo[a]pyrene, N-nitrosodimethylamine, and aflatoxin B1 into their ultimate carcinogenic metabolites that adduct DNA. These cells retain various enzymes involved in the metabolism of chemical carcinogens, including epoxide hydrolase, NADPH cytochrome P450 reductase, superoxide dismutase, catalase, glutathione S-transferases, and glutathione peroxidase.
