The knockout of genes is achieved by using gene editing tools to cut a known DNA sequence, causing DNA double-strand breaks and triggering the cell’s intrinsic repair mechanism. This leads to the deletion or insertion of bases during repair, resulting in a frameshift mutation and ultimately achieving the goal of gene knockout.Based on the slow virus gene editing tool, AcceGen has developed the TCR Knockout Jurkat Cell Line, confirmed the knockout at the DNA level through Sanger sequencing, and validated the knockout at the protein level through Western Blot and flow cytometry.
