Wnt proteins are secreted lipid-modified glycoproteins that can activate different intracellular signaling pathways. They regulate cell growth, function, differentiation, and cell death, which are essential for normal embryo development. Dysregulation of Wnt signaling can lead to various diseases, including cancer.The TCF LEF Reporter HEK-293 Cell Line is a luciferase reporter cell line constructed based on the Wnt signaling pathway. Wnt proteins activate the Frz receptor, leading to the translocation of β-catenin into the nucleus where it binds to TCF/LEF transcription factor binding sites and activates downstream pathways, resulting in the expression of luciferase. Luciferase readings represent the activation effect of the signaling pathway and can therefore be used for evaluating the in vitro effects of Wnt pathway-related drugs.
