The T84 Luciferase Reporter Cell Line is generated from the T84 colon epithelial cell line, which has been modified to constitutively express the luciferase gene. This allows for the real-time monitoring of cellular processes using luminescence as a reporter, making it a valuable tool for research in cellular signaling, drug discovery, and understanding colorectal carcinoma mechanisms.
The T84 Luciferase Reporter Cell Line should be cultured in DMEM/F12 medium supplemented with 10% fetal bovine serum (FBS) and 1% antibiotics-antimycotics. For selection and maintenance of the luciferase-expressing cells, 1 µg/mL puromycin can be added to the culture medium.
The T84 Luciferase Reporter Cell Line should be thawed rapidly in 37°C, ensuring that the vial is kept out of the water to avoid contamination. After thawing, these cells should be transferred to a centrifuge tube containing 5 mL of complete culture medium, centrifuged at 200×g for 5-10 minutes, and the supernatant discarded. The cell pellet should be resuspended in fresh medium and transferred to a new culture flask.
The T84 Luciferase Reporter Cell Line is particularly useful for studies involving gene expression regulation, cellular responses to pharmacological agents, and molecular pathways involved in colorectal cancer.
