The SK-BR-3 Luciferase Reporter Cell Line is used for studying eukaryotic gene expression and cell physiology. The luciferase gene system is applied due to its high sensitivity and success rate, enabling various applications such as promoter activity research, mammalian cell two-hybrid experiments, and in vivo imaging experiments.
Luciferase activity is validated using the Dual-Luciferase® Reporter Assay System Kit. The firefly luciferase reaction intensity is measured using a microplate reader, ensuring accurate detection of luciferase activity.
To thaw SK-BR-3-Luc cells:
Warm the complete culture medium to 37°C.
Transfer the cryopreserved vial from liquid nitrogen to a -80°C freezer briefly, then thaw it in a 37°C water bath.
After thawing, centrifuge the cells, discard the supernatant, and resuspend them in fresh medium before incubating them in a T25 flask in a 37°C, 5% CO₂ incubator.
The cells are shipped on dry ice. Upon receipt, they should be transferred immediately to a liquid nitrogen tank (-180°C) for long-term storage. The cells should not be stored at -20°C as this will not preserve them properly.
Each vial of the SK-BR-3-Luc cell line contains over 1×10⁶ cells with more than 90% viability. The cells are tested negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi.
