Description | The SK-BR-3 Luciferase Reporter Cell Line is a stable, clonal cell line derived from SK-BR-3 breast cancer cells, engineered to express the firefly luciferase gene. This cell line constitutively expresses luciferase, making it ideal for cell-based assays that require consistent and reliable signal detection. SK-BR-3 cells, originally isolated from a 43-year-old female adenocarcinoma patient, are known for overexpressing the HER2/c-erb-2 gene product. These cells are widely used in cancer research, particularly for studying HER2-positive breast cancer, and their luciferase expression allows for easy monitoring in experimental settings.
Why choose SK-BR-3 Luciferase Reporter Cell Line from AcceGen? SK-BR-3 Luciferase Reporter Cell Line from AcceGen has reliable quality. AcceGen ensures rigorous quality control, including mycoplasma-free certification and STR identification, providing consistent and authentic cells. With expertise in cell culture, this line is ideal for HER2-positive breast cancer research and drug testing.

Application | FOR RESEARCH USE ONLY The SK-BR-3 Luciferase Reporter Cell Line is a powerful tool for HER2-positive breast cancer research, particularly in exploring drug resistance mechanisms. Researchers can use these cells to investigate molecular pathways, providing critical insights into therapeutic targets and strategies to overcome resistance in this aggressive cancer subtype. Additionally, the luciferase expression allows for real-time monitoring of cell viability and drug efficacy. With its tumorigenic properties, the SK-BR-3 line is also useful for establishing adenocarcinoma models in nude mice, facilitating preclinical drug testing and the evaluation of antiproliferative activity of potential therapies.

Frequently Asked Questions
What is the purpose of using SK-BR-3 Luciferase Reporter Cell Line?
The SK-BR-3 Luciferase Reporter Cell Line is used for studying eukaryotic gene expression and cell physiology. The luciferase gene system is applied due to its high sensitivity and success rate, enabling various applications such as promoter activity research, mammalian cell two-hybrid experiments, and in vivo imaging experiments.
How is luciferase activity validated?
Luciferase activity is validated using the Dual-Luciferase® Reporter Assay System Kit. The firefly luciferase reaction intensity is measured using a microplate reader, ensuring accurate detection of luciferase activity.
What is the recommended procedure to thaw SK-BR-3-Luc cells?
To thaw SK-BR-3-Luc cells:
Warm the complete culture medium to 37°C.
Transfer the cryopreserved vial from liquid nitrogen to a -80°C freezer briefly, then thaw it in a 37°C water bath.
After thawing, centrifuge the cells, discard the supernatant, and resuspend them in fresh medium before incubating them in a T25 flask in a 37°C, 5% CO₂ incubator. What are the shipping and storage conditions for the SK-BR-3 Luciferase Reporter Cell Line?
The cells are shipped on dry ice. Upon receipt, they should be transferred immediately to a liquid nitrogen tank (-180°C) for long-term storage. The cells should not be stored at -20°C as this will not preserve them properly.
What quality control measures are implemented for the SK-BR-3-Luc Cell Line?
Each vial of the SK-BR-3-Luc cell line contains over 1×10⁶ cells with more than 90% viability. The cells are tested negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi.
