NUGC-4, also referred to as ‘Nagoya University-Gastric Cancer-4’, ‘NU-GC-4’, and ‘NUGC4’, is a human gastric cancer cell line established from the patient during surgery. This cell line was derived from signet ring cell carcinoma of human stomach at the metastatic paragastric lymph nodes. When cultured in vitro, NUGC-4 cells exhibit a spherical epithelial-like morphology and display characteristics of cell adhension, although a few free-floating cells may be also present. On average, the doubling time of NUGC-4 is about 30 hours, and it should be cultured in PRMI1640 with 10% FBS.

As a cancer cell line, NUGC-4 is widely used in scientific research, such as searching for key factors of the formation of human gastric cancer and targets for anti-cancer drugs. Epidermal growth factor (EGF) strongly activates the tyrosine phosphorylation in NUGC-4. Phosphatidylinositol 3-kinase (PI 3-kinase), which is mainly activated by tyrosine kinases, was precipitated with anti-phosphotyrosine antibodies in NUGC-4. Exogenous hepatocyte growth factor can stimulate the proliferation of NUGC-4, and MET tyrosine kinase inhibitors inhibited this proliferation of NUGC-4. Investigation of NUGC-4 cell line and the interactions with growth factors may provide the direction for identifying the signaling pathway of gastric cancer and screening of potential therapeutic targets.
