The NCI-N87 Luciferase Reporter Cell Line is derived from the NCI-N87 cell line, originally isolated from a human gastric carcinoma. This reporter cell line has been engineered to stably express the luciferase gene, enabling the real-time tracking of cellular processes using luminescent reporting. This feature makes it particularly useful for research in gastric cancer biology, drug response studies, and other applications where bioluminescent imaging is beneficial.
The NCI-N87 Luciferase Reporter Cell Line should be cultured in RPMI 1640 medium supplemented with 10% fetal bovine serum (FBS) and 1% antibiotics-antimycotics. To maintain selective pressure and ensure stable luciferase expression, 0.5 µg/mL puromycin should be added to the culture medium.
Upon receipt, the cells should be rapidly thawed in a 37°C water bath for approximately 2 minutes to minimize the risk of ice crystal formation and cellular damage. After thawing, cells should be decontaminated using 70% ethanol, transferred to a centrifuge tube containing complete culture medium, centrifuged at 200×g for 5-10 minutes, and the supernatant discarded. The cell pellet should be resuspended in fresh medium and transferred to a new culture flask.
The NCI-N87 Luciferase Reporter Cell Line is extensively used in gastric cancer research to study gene expression, signal transduction pathways, and cellular responses to therapeutic agents. The luciferase expression allows for non-invasive tracking of tumor cell behavior in vivo and is ideal for assessing the efficacy of anti-cancer drugs in preclinical models.
