MOLM-13 is a human leukemia cell line derived from the peripheral blood of a patient with acute myeloid leukemia(AML), FAB M5a, at relapse. The immunotype of MOLM-13 includes CD34+, CD13−, CD14−, CD15+, CD33+, CD3−, CD4+, cy CD68+, HLA-DR−. After stimulated by INF-γ alone, or in combination with TNF-α, MOLM-13 can differentiate into macrophage-like morphology and express upregulated myelomonocyte-associated antigens. Internal tandem duplication of the receptor tyrosine kinase FLT3 gene is a unique feature of MOLM-13, compared to other leukemia cell lines. MOLM-13 can be cultured in RPMI1640 supplemented with 10% FBS, and the split ratio used to passage cells is 1:2 or 1:3 every 2-3 days.

MOLM-13 is a useful in vitro model to investigate monocytic differentiation and leukemogenesis. For example, used in reverse transcriptase polymerase chain reaction (RT-PCR) analysis, MOLM-13 was shown to contain the mRNA of the MLL-AF9 fusion gene, which is a kind of MLL gene rearrangement and related to the leukemia development. MOLM-13 can be used to identify the genetic file of human leukemia and search for more genetic marker, such as MLL-AF9 for the diagnosis and prognosis. MLL-rearranged leukemia is hard to treat, MOLM-13 may be used in more scientific research to discover the leukemia mechanisms based on MLL fusions.
