Growth differentiation factor 15 (GDF15) is a distal member of the transforming growth factor (TGF)-b family, existing as a secreted protein circulating as a 25 kDa dimer. It belongs to the transforming growth factor β superfamily. Normally, GDF-15 is expressed at low levels in most organs but upregulated during organ damage such as in the liver, kidneys, heart, and lungs.Growth factor receptor bound protein 2 like (GFRAL), derived from glial-cell line-derived neurotrophic factor (GDNF) family, is the sole receptor for GDF15. GDF15 binds to GFRAL with high affinity, selectively interacts with GFRAL but not with other members of the GDNF receptor family. Upon binding with GDF15, GFRAL forms a complex with the co-receptor RET, leading to typical cell signaling seen in the GDNF family and associated with phosphorylation of RET, AKT, ERK, and PLC-γ1.The Mouse_GDF15 Reporter 293 Cell Line is a reporter gene cell line constructed based on the RET-ERK signaling pathway. Upon binding of GDF15 to its receptor GFRAL and then forming a complex with the co-receptor RET, RET is phosphorylated, subsequently leading to ERK phosphorylation and activation of ELK1. ELK1 translocates to the nucleus to activate the expression of luciferase. The measurement of luciferase activity represents the activation effect of the signaling pathway, making it useful for evaluating the in vitro effects of GDF15-related drugs.
