IL-36 is a member of the IL1 cytokine family. Currently, four subtypes have been identified: IL-36α (IL-1F6), IL-36β (IL-1F8), IL-36γ (IL-1F9), and IL-36Rα (IL-1F5); the first three are agonists of IL-36 receptors, promoting inflammation, while IL-36Rα is a receptor antagonist, countering inflammation. The active forms of IL-36α, IL-36β, and IL-36γ proteins are produced through cleavage. By binding to IL-36 receptors and activating signaling pathways, they release cytokines, chemokines, and promote inflammation. The active form of IL-36Rα, unable to bind to IL-36 receptors and initiate signaling, exhibits only anti-inflammatory effects.IL-36 Reporter 293 Cell line is a stable reporter cell line expressing IL-36R, IL-1RAcP, and Luciferase genes. When IL-36α/β/γ bind to the IL-36R and IL-1RAcP receptors, the signaling pathway is activated, leading to the expression of luciferase. Luciferase readings represent the activation effect of the signaling pathway and can therefore be used to evaluate the in vitro effects of IL-36-related drugs.
