Interleukin-2 (IL-2) is a cytokine responsible for regulating the immune activity of white blood cells, playing a crucial role in the immune system. IL-2 binds to IL-2 receptors on the surface of lymphocytes and transmits signals into the cell. The IL-2 receptor is composed of α, β, and γ chains, forming a heterotrimer (IL-2Rα, IL-2Rβ, IL-2Rγ). IL-2 activates JAK signal transduction and phosphorylation of Signal Transducer and Activator of Transcription (STAT) pathways, leading to the translocation of phosphorylated STAT dimers or tetramers to the nucleus. The IL2 Reporter Cell Line is a reporter gene cell line constructed based on the JAK1/3-STATs signaling pathway. When IL-2 binds to its receptor, the receptor trimer and JAK1/3 are phosphorylated, further mediating the phosphorylation of STATs and the translocation of STAT dimers or tetramers to the cell nucleus. This process activates the expression of luciferase. Luciferase readings represent the activation effect of the signaling pathway, making it suitable for evaluating the in vitro effects of IL-2-related drugs.
