Interleukin-17 Family. This family of cytokines is named interleukin-17A to interleukin-17F. Their receptor family includes interleukin-17 receptor A to interleukin-17 receptor E. These interleukin-17 cytokines can bind to their corresponding receptor members to mediate different inflammatory responses.The most representative member of this family, interleukin-17A (IL-17A), is secreted by lymphocytes that migrate to infected or damaged areas in the body. IL-17A induces the expression of inflammatory and chemotactic factors, recruiting more immune cells to the site of inflammation, exacerbating the expression of inflammatory factors in the body. Elevated levels of IL-17A worsen the pathological development of diseases. However, IL-17A also induces the expression of tissue repair-related factors, accelerating the body’s recovery.Although IL-17A plays a role in expanding the immune defense response and protecting the body during infection and tissue repair processes, it is overexpressed in many autoimmune disease patients and cancer patients due to its pro-inflammatory effects. Animal experiments have shown that the deficiency of IL-17A or neutralization of IL-17A with antibodies can effectively suppress the development of various autoimmune diseases.The IL-17 receptor family consists of 5 members: IL-17RA to IL-17RE. The receptor for IL-17A is a heterodimeric complex formed by IL-17RA and IL-17RC. So far, all IL-17 receptors use Act1 as the bridging molecule for downstream signal transduction.The IL17A Reporter 293 Cell Line is a luciferase reporter cell line based on the IL-17 signaling pathway. When IL-17A binds to the IL-17RA-RC receptor, it recruits Act1, which then binds to TRAF6, further activating the expression of luciferase. The luciferase readout represents the activation effect of the signaling pathway, making it useful for evaluating the in vitro effects of IL-17A-related drugs.
