Interleukin-15 (IL-15) is a pro-inflammatory protein that plays a role in the activation of neutrophils, dendritic cells, and macrophages, and is essential for the development and survival of NK cells and CD8+ T cells.IL-15 binds to the IL-15Rα receptor, then is presented to surrounding cells bearing the IL-15Rβγ complex on their surface, where it binds to IL-15Rβ and IL-15Rγ to form a heterotrimeric receptor complex. IL-15 triggers JAK signaling and phosphorylation of signal transducer and activator of transcription (STAT) pathway, leading to the translocation of phosphorylated STATs dimers or tetramers to the nucleus.The IL15 Reporter Cell Line is a luciferase reporter cell line constructed based on the JAK1/3-STATs signaling pathway. When IL-15 binds to its receptor, the receptor trimer and JAK1/3 are phosphorylated, subsequently mediating the phosphorylation of STATs, causing the translocation of STATs dimers or tetramers to the nucleus. This activation leads to the expression of luciferase. Luciferase readings represent the activation effect of the signaling pathway and can be used for in vitro evaluation of IL-15 related drugs.
