Interleukin-12 (IL-12) is a cytokine produced naturally in response to antigen stimulation by dendritic cells, macrophages, neutrophils, and human B lymphoblastoid cells (NC-37). It belongs to the interleukin 12 family, which includes unique heterodimeric cell factors such as IL-12, IL-23, IL-27, and IL-35.IL-12 is a heterodimeric cytokine encoded by two independent genes, IL-12A (p35) and IL-12B (p40).The IL-12 receptor is a heterodimer composed of IL-12Rβ1 and IL-12Rβ2. Once combined on the cell membrane, IL-12Rβ1 binds TYK2, and IL-12Rβ2 binds Jak2. The formation of the receptor complex promotes the phosphorylation of receptor-associated kinases JAK2 and TYK2, as well as phosphorylation of the signal transduction binding site on the cytoplasmic tail of IL-12Rβ2. This phosphorylation of the IL-12Rβ2 binding site attracts STATs, which are then phosphorylated by JAK2. Phosphorylated STATs dimerize and translocate into the nucleus.The IL12 Reporter 293 Cell Line is a reporter gene cell line constructed based on the JAK-STAT signaling pathway. When IL-12 binds to the receptor to form a complex, the receptor complex is phosphorylated by JAK1 and Tyk2. This ultimately leads to the phosphorylation and dimerization of STATs and subsequent translocation, activating the expression of luciferase. Luciferase readings represent the activation effect of the signaling pathways and can be used for the in vitro evaluation of IL-12 related drugs.
