IL-27 is a heterodimer composed of Epstein-Barr virus-induced gene 3 (EBI3) subunit and p28 subunit, which are associated with p40 and p35, respectively. IL-27 binds to a unique receptor subunit called WSX-1/TCCR (IL-27RA), which is related to the common signaling subunit gp130 used by the IL-6 cytokine family. IL-27RA is highly expressed on T cells and NK cells. Binding of IL-27 to the receptor leads to activation of JAK, which then activates STAT1-5.IL-27 is produced by activated DCs, macrophages, and microglia cells, exhibiting pro-inflammatory and anti-inflammatory effects depending on the cellular environment. In terms of pro-inflammation, IL-27 promotes differentiation of T cells into Th1 lineage by inducing the transcription factor T-bet. Additionally, IL-27 inhibits Th2 cell differentiation, thereby suppressing secretion of Th2 cell cytokines like IL-4, -5, and -10. In terms of anti-inflammation, IL-27 inhibits differentiation of Th17 cells and secretion of IL-17A. Furthermore, IL-27 induces the production of anti-inflammatory cytokine IL-10.IL-27 Reporter 293 Cell Line is a luciferase reporter cell line. When IL-27 binds to the receptor, downstream signaling pathways are activated, leading to expression of luciferase. Luciferase readings represent the activation effect of the signaling pathway, making it suitable for evaluating the in vitro effects of IL-27-related drugs.
