The liver fulfills many vital processes in mammals. It is the central organ of energy metabolism, biotransformation of xenobiotics, and synthesis of plasma proteins under physiological and pathophysiological conditions. Primary human hepatocyte culture is an important experimental model for the study of liver specific function. Propagation of human hepatocytes is critical for gene therapy, cell […]

Description

Human Hepatocytes (HH) are hepatic cells isolated from the whole human liver, making up 80% of the liver’s mass. Liver hepatocytes are multifunctional. Hepatocyte involves in the production and synthesis of functional proteins that participate in energy metabolism, biotransformation, detoxification, etc. Hepatocytes secrete many materials including structural protein and intracellular enzymes, cholesterol, bile, detoxify substances. Understanding the molecular mechanisms of the liver is beneficial to elucidate new therapies for treatment of hepatic disease.

Primary hepatocyte cultures are unable to expand, which is challenging for long-term research on understanding the regulation of human hepatocytes.

Why Choose Human Hepatocytes from AcceGen?

AcceGen cryopreserves our Human Hepatocytes for about 1 x 10^6 cells/mL, characterized by immunofluorescence with specific antibodies against albumin, cytokeratin-18 and vimentin.

Expanding or long-term culturing is not recommended, as the cells do not proliferate in culture.

Immortalized Human Hepatocytes are also available in AcceGen.

Frequently Asked Questions

  • What are Human Hepatocytes, and how are they obtained?

Human Hepatocytes are the main functional cells of the liver, responsible for various metabolic processes, detoxification, and protein synthesis. They are isolated from donor’s liver tissue through enzymatic digestion and are either freshly isolated or cryopreserved for research use.

  • What are the primary applications of Human Hepatocytes in research?

Human Hepatocytes are widely used in drug metabolism studies, toxicology testing, liver disease research, pharmacokinetic studies, and tissue engineering. They serve as a critical model for understanding liver function and drug interactions.

  • How do Human Hepatocytes differ from hepatoma cell lines?

Human Hepatocytes are primary cells with normal liver cell functions, making them more physiologically relevant for certain studies. In contrast, hepatoma cell lines, such as HepG2, are cancer-derived and may not accurately represent normal liver function, particularly in drug metabolism and toxicity studies.

  • Can Human Hepatocytes be passaged in culture?

No, Human Hepatocytes cannot be passaged in culture like immortalized cell lines. They are primary cells, meaning they have a limited lifespan and do not proliferate in culture. After a few days in culture, hepatocytes will begin to lose their specific liver functions and eventually undergo senescence. For extended studies, it is often necessary to use fresh or cryopreserved hepatocytes for each experiment rather than attempting to passage them.

  • What types of Human Hepatocyte products are available?

We offer both suspension and Platable Human Hepatocytes in various formats and quantities. Please contact our sales team for detailed information on the specific options available.
