Growth differentiation factor 15 (GDF15) is a distantly related member of the transforming growth factor (TGF)-β family, circulating as a secreted protein in the form of a 25 kDa dimer. It belongs to the transforming growth factor β superfamily. Normally, GDF-15 is lowly expressed in most organs, but its expression is upregulated in organs like liver, kidney, heart, and lungs in the presence of injury. Growth differentiation factor receptor alpha-like (GFRAL), derived from the glial-cell line-derived neurotrophic factor (GDNF) family, is the sole receptor for GDF15. GDF15 binds to GFRAL with high affinity, exclusively interacting with GFRAL and forming a complex with a co-receptor RET after binding GDF15, typical of GDNF family cell signaling, associated with phosphorylation of RET, AKT, ERK, and PLC-γ1.The GDF15 Reporter 293 Cell Line is a luciferase reporter cell line constructed based on the RET-ERK signaling pathway. When GDF15 binds to its receptor GFRAL and then forms a complex with co-receptor RET, it induces the phosphorylation of RET, further leading to ERK phosphorylation and activation of ELK1. ELK1 translocates into the nucleus activating luciferase expression. Luciferase readings represent the activation effect of the signaling pathway and can be used for in vitro evaluation of GDF15-related drug effects.
