The epidermal growth factor receptor (EGFR) is a type I transmembrane protein that belongs to the receptor of the extracellular protein ligand epidermal growth factor family (EGF family). When EGFR binds to its ligand, it induces receptor heterodimerization and autophosphorylation on cytoplasmic residues, activating intracellular signaling pathways to promote cell growth, division, proliferation, and cell survival. Inhibiting EGFR signal transduction can be achieved by blocking EGFR binding sites or inhibiting intracellular tyrosine kinase activity.Mutations in EGFR can lead to sustained activation of the EGFR signaling pathway in the absence of ligand binding, resulting in abnormal cell proliferation. EGFR mutations have been identified in various tumors, such as non-small cell lung cancer, breast cancer, colon cancer, and head and neck squamous cell carcinoma (HNSCC).EGFR Reporter Cell Line is a Luciferase reporter gene cell line that, upon binding of EGF to the receptor, activates downstream signaling pathways, thereby activating the expression of firefly luciferase. The luciferase readout reflects the activation effect of the signaling pathway and can be used for in vitro evaluation of EGFR-related drug effects.
