Description

The calcitonin gene-related peptide (CGRP) family includes calcitonin, amylin, CGRP, and adrenomedullin. Among them, human α-CGRP and β-CGRP proteins differ by three amino acids.Receptors of the CGRP family consist of two subtypes: calcitonin receptors (CALCR, CTR) binding to CT, and calcitonin receptor-like receptor (CRLR, CLR, CALCRL). The CGRP receptor comprises calcitonin receptor-like receptor, receptor activity-modifying protein (RAMP1), and CGRP receptor component protein (CRCP). RAMP1 is essential for transporting the CGRP receptor to the membrane and coupling with CGRP agonists. CRCP is involved in the interaction between the CGRP receptor and G protein. Upon ligand binding, the complex interacts with the G protein, leading to adenylate cyclase activation, cAMP production, and the stimulation of cAMP-dependent signaling pathways, ultimately inducing gene expression.Preclinical evidence suggests that during migraines, sensory neurons activated in the trigeminal ganglion release CGRP from nerve terminals in the meninges. Released CGRP binds and activates the CGRP receptor, causing vasodilation and plasma extravasation. Therefore, CGRP antagonists are becoming prominent in migraine treatment.CALCRL RAMP1 Reporter Cell Line is a luciferase reporter cell line based on the cAMP signaling pathway. When CGRP binds to CALCRL/RAMP1, it activates the expression of luciferase by stimulating the cAMP signaling pathway. Luciferase readings reflect the activation effect of the signaling pathway, making it suitable for evaluating the in vitro effects of CGRP-related drugs.
