Cre Expressing Cells

Cre recombinase is a highly versatile enzyme that plays a crucial role in site-specific DNA recombination. Originating from the P1 bacteriophage, Cre recombinase mediates recombination between two specific DNA recognition sites known as loxP sites. Each loxP site consists of a 34 base pair sequence, composed of two 13 bp palindromic sequences flanking an asymmetric 8 bp spacer region. Depending on the orientation and position of these loxP sites, Cre recombinase can either excise the DNA between them, resulting in a circular plasmid containing the excised DNA, or fuse two DNA fragments. This reaction operates at equilibrium, does not require additional cofactors, and functions both in vitro and in vivo across a wide range of organisms, making the Cre/loxP system a powerful tool in biotechnology.
