Fluorescent proteins (FPs) are naturally occurring molecules found in certain organisms, known for their remarkable ability to emit fluorescence when expressed in prokaryotic or eukaryotic cells. This fluorescence occurs without the need for additional enzymes or cofactors, aside from molecular oxygen. Scientists have harnessed this unique characteristic to develop powerful tools for biological research, particularly in imaging experiments. The first fluorescent protein to be discovered was the Green Fluorescent Protein (GFP). GFP is remarkable because it can produce a highly visible and efficiently emitting fluorophore internally, making it invaluable as a marker for gene expression and protein targeting in intact cells and organisms. Advances in mutagenesis and the engineering of GFP into chimeric proteins have expanded its utility, leading to innovations in physiological indicators, biosensors, and photochemical memories. Following GFP, Red Fluorescent Protein (RFP) was cloned. RFP differs significantly from GFP in its spectral properties, as it emits red fluorescence. Over the years, extensive research has focused on developing RFP mutants to enhance brightness, photostability, maturation efficiency, pH stability, and reduce cytotoxicity. These improvements have significantly increased the effectiveness of RFPs in live-cell microscopy, making them indispensable tools in modern biological research.
