Caco-2 is a human epithelial cell line derived from colon carcinoma. It exhibits characteristics similar to normal enterocytes when grown on plastic dishes or nitrocellulose filters. Caco-2 cells have the ability to undergo differentiation and form a monolayer of cells that exhibit numerous characteristics commonly seen in absorptive enterocytes, including the presence of a brush border layer resembling that of the small intestine. Over time, these cells show increased in alkaline phosphatase activity in the culture, highlighting their epithelial cell nature. Notably, Caco-2 cells possess a specific genetic mutation (heterozygous G-to-C missense transversion) in codon 245 of exon 5. This mutation results in a substitution of glycine to alanine, potentially leading to impaired target recognition by protein kinase enzymes and altered accessibility for phosphorylation at that site.
