RAW-264.7 cells, originating from a BALB/c mouse tumor transformed by the Abelson leukemia virus, are a semi-adherent macrophage-like cell line highly regarded for their rapid proliferation and simple handling. With a doubling time spanning from 11 to 30 hours, these cells enter their maintenance phase within approximately 24 hours of adhering to the culture dish surface, necessitating media replenishment every 2-3 days but no later than 4 days. Typically measuring 10-20μm in diameter, RAW264.7 cells showcase essential macrophage functions, including pinocytosis and phagocytosis. As immortal cancer cells, they contribute to regulating bone remodeling without being osteoclasts. The RAW-264.7 cell line serves as a valuable model for mouse macrophages in the study of cellular responses to microbes and their products, making it an essential tool for immunology and microbiology research. These cells are extensively employed in investigations related to osteoclasts, as they can differentiate into osteoclast-like cells (OCLs), enabling the exploration of underlying mechanisms and signal pathways governing precursor cell migration towards bone. Additionally, RAW-264.7 cells are a preferred choice as a transfection host due to their suitability, further enhancing their utility in various experimental contexts.
The RAW 264.7 cells are monocyte/macrophage-like cells. The normal cell morphology generally showed a round and smooth form. A small percentage (<10%) of spindle-shaped or polygonal differentiated cells is considered normal.
Please subculture when density reached 70-80%. Cells should not overgrow and become confluent as this can lead to loss of the flattened adherent cell characteristic, prone to differentiate small tentacles.
Cells are semi-adherent, it is recommended to use cell scrapers to gently remove attached cells. Do not use trypsin. Please seed the undifferentiated cells only.
The RAW-264.7 cell seeding is approximate 2-4×10,000 cells/cm². Unlike other cell lines, RAW-264.7 cells do not connect when density is low, instead, they tend to grow in clusters, distributed in patches in the culture vessel.
Induction time and cell density are two key factors related to the differentiation of RAW264.7 cells. It is hard to avoid the differentiation and please control the differentiation ratio into a certain range. Please culture the RAW-264.7 cell line carefully.
Also, RAW264.7 cells might act as osteoclast progenitors and could differentiate to osteoclasts in response to stimulation substances like LPS.
The growth media should be refreshed every 2-3 days, mostly not over 4 days. The average doubling time of RAW cells is 15 hours.
