CTX TNA2 represents a type I astrocyte cell line, derived from primary cultures of type 1 astrocytes sourced from the frontal cortex tissue of neonatal rats. Following an initial plating phase, these cultures underwent transfection with a DNA construct harboring the oncogenic early region of SV40, which was placed under the transcriptional regulation of the human GFAP promoter (pGFA-SV-Tt), along with pPGK-neo featuring the murine phosphoglycerate kinase gene promoter. As a result, CTX TNA2 cells have retained key characteristics consistent with the type 1 astrocyte phenotype. Notably, around 20% of these cells exhibit immunoreactivity for glial fibrillary acidic protein (GFAP), indicating their astrocytic identity. Additionally, they possess a high-affinity gamma-aminobutyric acid (GABA) uptake mechanism, which is susceptible to inhibition by beta-alanine. In terms of protein production, CTX TNA2 cells synthesize alpha 2 macroglobulin at levels similar to those found in primary astrocytes but exhibit markedly reduced transferrin production. Immunostaining analysis further confirms the presence of SV40 T-antigen within the nuclei of over 95% of these cells.
